In this manuscript, the authors come up with a method to enhance the recovery rates of Cryptosporidium Oocysts from contaminated water samples. They spike water with known quantity of the oocysts and trap it in filter. The method described details the efforts to get the oocysts out of the filter for detection. Megasonic based approaches have been already used for this purpose, so it is unclear how using different set of filters will make a difference. What is the rationale to spike water with 100 oocysts, how many did they recover from the filter and how was this assessed? These details are important, but not explicated. It is also important to show an effect of different megasonic output setting on recovery rates.
Nevertheless, this method has previously been used for detection of Cryptosporidium, hence the novelty is not clear.
