- Considering that the authors are presenting what could be a new property of EVs, a major conceptual problem is that these membranes could be pieces of plasma membrane that are shed from cells, and not true EVs. Can the authors separate these, for example, by staining with markers enriched in exosomes but not the plasma membrane?
- Quantitation of what is often qualitatively described is needed. For example, only a “small fraction” shows reshaping, but what is the percentage? Is it the 1.3% that contains actin? Does the average size of the fraction that shows morphological changes in light microscopy correspond to the electron microscopy, and do the sizes of the ones that contain actin correlate to these? Are the sizes that undergo reshaping comparable in microvesicles and exosomes? It seems as if the authors have all these data, and considering their small sample sizes, they could get these numbers in a few days.
