A few points.
Throughout, human versions of protein names should be capitalised.
Expression constructs are not stated to have been sequence verified.
In the version I have to assess I cannot find legends for any of the figures other than figure A.
I also cannot find the description/legend for supplemementary data figure F.
N numbers need to be stated for each experiment throughout the manuscript and it needs to be stated what each n is, technical or biological replicate, field of view etc.
Quantification of immunostaining is not acceptable in this format. There must be much more detail about how the quantification was achieved and controlled. The n=4 is difficult to understand (maybe again because the legend is missing), is this 4 fields of view? In any case n=4 will never be sufficient for quantifying immunostaining. There should also be control cells shown without primary antibody/secondary antibody etc. There is no specific information about which antibodies and dilutions are used for this set of experiments. Usually dilutions and even antibodies are different between western blotting and immunostaining.
Addressing these issues could make this a better quality manuscript from a technical point of view.
